Abstract or lay summary: The goal of the research described in this proposal is to bring a human stem cell derived product, SD56, to IND filing for treatment of motor sequelae of sub-cortical stroke. SD56 is a neural precursor cell line that is amendable to large-scale production sufficient for an appropriate quality assurance program that is required for multi-center clinical trials. SD56 engraftment into the peri-infarct area of a rodent stroke model has been shown to improve neurologic outcome. In order to develop assays for the potency of SD56, we have evaluated the mechanisms involved in the SD56-mediated improvement of recovery from stroke. Our research has identified endogenous repair mechanisms enhanced by SD56 transplantation by secretion of trophic factors. To increase the survival of transplanted cells, as well as their ability to produce/deliver trophic factors, we are injecting the cells in a hydrogel matrix, which may lead to greatly increased efficacy. We have designed a research program that encompasses the development and manufacture of the SD56 line, the pharmacology and toxicology of SD56 transplanted with and without hydrogel all within a framework of operation that will meet regulatory requirements for IND filing.
